What is Vosevi (Sofosbuvir - velpatasvir - voxilaprevir)?

Vosevi is a once-daily single-tablet regimen combining three direct-acting antiviral agents used to treat chronic hepatitis C infection across all major genotypes (1–6), particularly in patients who have been previously treated with certain other hepatitis C regimens. Each component targets a different protein the hepatitis C virus needs to replicate, making it extremely difficult for the virus to develop resistance. Treatment is typically 8 or 12 weeks in duration. Cure rates—defined as undetectable virus 12 weeks after completing treatment—are consistently above 95%, representing a remarkable transformation in the treatment of what was once a lifelong chronic illness.

Triple direct-acting antiviral mechanism

Sofosbuvir inhibits NS5B polymerase, velpatasvir blocks NS5A protein, and voxilaprevir inhibits NS3/4A protease—three essential viral enzymes targeted simultaneously, making resistance essentially impossible with consistent adherence.

Pan-genotypic coverage

Effective against all major hepatitis C genotypes (1 through 6) in a single fixed-dose regimen, eliminating the need for genotype-specific treatment selection and simplifying care across diverse patient populations.

High cure rate re-treatment option

Specifically developed for patients who have failed previous NS5A inhibitor-containing regimens—providing a rescue option with sustained virologic response (cure) rates above 95% even in this difficult-to-treat population.

Frequently Asked Questions

What is Vosevi used for?

Vosevi is a complete once-daily hepatitis C treatment approved for adults with all major HCV genotypes — particularly those who have previously been treated with NS5A inhibitor-containing regimens and need a rescue option.

How effective is Vosevi at curing hepatitis C?

Sustained virologic response (cure) rates with Vosevi consistently exceed 95% — even in patients who failed previous treatment — representing one of the highest cure rates in the history of infectious disease treatment.

How long is the Vosevi treatment course?

Most patients complete treatment in 8 or 12 weeks — determined by HCV genotype, treatment history, and whether cirrhosis is present. Your clinician will specify the appropriate duration based on your situation.

What side effects might I experience?

Headache, fatigue, nausea, and diarrhea are most commonly reported — generally mild. Vosevi can reactivate hepatitis B in patients with prior HBV infection, so HBV testing is required before starting treatment.

Will I need follow-up testing after completing Vosevi?

Yes. A hepatitis C viral load test is performed 12 weeks after completing treatment to confirm sustained virologic response — the definition of cure. A negative result at this point means the infection has been eliminated.
