What is Trimipramine?

Trimipramine is a tricyclic antidepressant used to treat depression, and at lower doses is sometimes used for insomnia and anxiety. It works by blocking the reuptake of serotonin and norepinephrine while also blocking histamine and other receptors—producing both antidepressant and sedating effects. Unlike most antidepressants, trimipramine has minimal effects on REM sleep architecture, making it particularly notable for patients where preserving normal sleep structure is a consideration. It's taken once daily, typically at bedtime given its sedating properties.

Serotonin and norepinephrine reuptake inhibition

Blocks the reuptake of both serotonin and norepinephrine, increasing their availability in the brain to regulate mood, reduce anxiety, and support emotional stability over time with consistent use.

Sedating antihistamine activity

Potent H1 histamine receptor blockade produces significant sedation—making trimipramine useful for depression accompanied by insomnia, and sometimes prescribed at lower doses primarily for sleep difficulties.

REM sleep preservation

Unlike many antidepressants that suppress REM sleep, trimipramine has minimal effects on REM architecture—a meaningful distinction for patients who experience REM-related symptoms or for whom normal sleep structure is a treatment priority.
