What is Prucalopride?

Prucalopride is a prescription medication used to treat chronic idiopathic constipation in adults—constipation with no identifiable underlying cause that persists despite standard measures. It works by selectively activating serotonin 5-HT4 receptors throughout the gastrointestinal tract, which stimulates the coordinated muscle contractions that propel food and stool through the colon. Unlike laxatives that draw water into the bowel, Prucalopride works on the underlying motility mechanism—improving the colon's ability to move stool forward on its own. It's taken once daily.

Selective 5-HT4 receptor agonism

Activates serotonin 5-HT4 receptors along the gastrointestinal tract, stimulating the enteric nervous system to increase the frequency and coordination of colonic peristaltic contractions that move stool through the bowel.

Prokinetic mechanism

Rather than drawing water into the colon like osmotic laxatives, Prucalopride restores the colon's own motility—addressing the underlying dysfunction in how the colon moves rather than compensating for it with external agents.
