What is Memantine?

Memantine is a prescription medication used to treat moderate to severe Alzheimer's disease. It works through a different mechanism than cholinesterase inhibitors like donepezil—rather than boosting acetylcholine, it regulates glutamate activity in the brain, which becomes overactive in Alzheimer's and contributes to neuronal damage and cognitive decline. By moderating this excess glutamate signaling, Memantine helps preserve cognitive function, slow symptom progression, and maintain daily functional abilities for longer. It's taken once or twice daily and is often used alongside a cholinesterase inhibitor for a complementary dual approach.

NMDA receptor modulation

Blocks NMDA glutamate receptors in a voltage-dependent, use-dependent manner—reducing the pathological glutamate overactivation that contributes to excitotoxic neuronal damage in Alzheimer's disease while allowing normal glutamate signaling to continue.

Cognitive function preservation

By reducing excitotoxic damage, Memantine helps slow the decline in memory, thinking, and daily function associated with moderate-to-severe Alzheimer's—not reversing the disease, but slowing its progression.

Complementary to cholinesterase inhibitors

Works through a completely different mechanism than donepezil, rivastigmine, and galantamine—making it an effective add-on therapy that targets Alzheimer's pathophysiology through two independent pharmacological pathways simultaneously.
