What is Malarone?

Malarone is a prescription antimalarial medication combining atovaquone and proguanil, used to both prevent and treat malaria caused by Plasmodium falciparum—the most dangerous malaria species. For prevention, it's taken once daily starting 1–2 days before travel to a malaria-endemic area and continuing for 7 days after return—a shorter pre- and post-travel window than some other antimalarials. The combination of two drugs with complementary mechanisms makes it highly effective and harder for the malaria parasite to develop resistance against. It's well-tolerated and a preferred choice for many travelers.

Dual antimalarial mechanisms

Atovaquone disrupts mitochondrial electron transport in malaria parasites, collapsing their energy production, while proguanil's active metabolite inhibits dihydrofolate reductase—blocking parasite DNA synthesis through a second independent pathway.

Targeting two essential parasite processes simultaneously makes it significantly harder for Plasmodium falciparum to develop drug resistance compared to single-agent antimalarials.
