What is Denavir (Penciclovir)?

Denavir is a prescription antiviral cream used to treat recurrent cold sores (herpes labialis) caused by herpes simplex virus type 1. Applied directly to the affected area, it works by blocking the enzyme the virus needs to replicate its DNA—limiting the virus's ability to spread and shortening the duration and severity of the outbreak. It's applied every 2 hours while awake for 4 days, starting at the earliest sign of a cold sore such as tingling, itching, or redness. Early application gives the best chance of reducing the outbreak's impact.

Viral DNA polymerase inhibition

Penciclovir is activated by a viral enzyme (thymidine kinase) inside HSV-infected cells and then blocks the viral DNA polymerase, preventing the virus from replicating its genetic material and producing new viral particles.

Virus-selective activation

Penciclovir is preferentially activated inside herpes-infected cells rather than healthy tissue—concentrating its antiviral effect at the site of active viral replication while minimizing effects on surrounding uninfected skin.

Early-use outbreak shortening

Clinical studies show meaningful reductions in healing time, pain duration, and viral shedding when applied at the first sign of a cold sore—making prompt application at the tingle or itch stage essential for maximum benefit.

Frequently Asked Questions

When should I start applying Denavir?

Apply Denavir at the very first sign of a cold sore — the tingling, itching, or burning that signals one is developing. Starting early, before the blister appears, gives the best chance of reducing severity and duration.

How often do I need to apply it?

Denavir is applied every 2 hours while awake for 4 days — starting at the first sign of the outbreak. Consistent application throughout the day is important for the medication to be effective.

Can Denavir prevent cold sores from developing?

No. Denavir is a treatment for active outbreaks — it cannot prevent them from occurring. Antiviral tablets like valacyclovir taken daily are used for suppressive therapy to reduce outbreak frequency.

What side effects might I experience?

Local reactions at the application site — redness, stinging, and a temporary numbing or altered sensation — are the most commonly reported effects. Headache has also been reported. Serious side effects are uncommon with topical use.

Can I apply Denavir inside my mouth?

No. Denavir is for external cold sore use on the lips and face only — it should not be applied inside the mouth, near the eyes, or on mucous membranes. Wash hands thoroughly after each application.
