What is Clinoril (sulindac)?

Clinoril is a non-steroidal anti-inflammatory drug (NSAID) used to treat pain, inflammation, and stiffness associated with osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, acute gout, and other musculoskeletal conditions. Like other NSAIDs, it works by inhibiting COX-1 and COX-2 enzymes to reduce prostaglandin production—decreasing inflammation and pain. Sulindac is a prodrug, meaning it's converted to its active form in the body, which may offer some GI advantages over other NSAIDs. It's taken twice daily and, like all NSAIDs, should be used at the lowest effective dose for the shortest duration.

Prostaglandin synthesis inhibition

Blocks both COX-1 and COX-2 enzymes, reducing the production of prostaglandins that drive pain, swelling, heat, and stiffness in arthritis and other inflammatory musculoskeletal conditions.

Prodrug conversion

Sulindac itself is pharmacologically inactive and is converted to the active sulfide form in the body—this prodrug design may reduce direct GI irritation compared to NSAIDs that are immediately active in the stomach.
