What is Benlysta (Belimumab)?

Benlysta is a biologic medication approved for the treatment of active systemic lupus erythematosus (SLE) and lupus nephritis in adults and children aged 5 and older. It's the first new lupus treatment approved in more than 50 years. Belimumab works by targeting BLyS (B lymphocyte stimulator), a protein that promotes the survival of autoreactive B cells—the immune cells that produce the self-attacking antibodies responsible for lupus organ damage. By blocking BLyS, Benlysta reduces the survival of these harmful B cells, lowering autoantibody levels and reducing flares and organ damage over time. It's given as a monthly IV infusion or weekly subcutaneous injection.

BLyS inhibition

Binds to and blocks B lymphocyte stimulator (BLyS), a survival factor for autoreactive B cells—reducing the population of B cells that produce the anti-dsDNA and other autoantibodies that drive lupus inflammation and organ damage.

Autoantibody reduction

By limiting the survival of BLyS-dependent autoreactive B cells, Benlysta lowers circulating autoantibody levels over time, reducing the immune complex deposition that causes lupus nephritis, joint inflammation, and other organ involvement.

Flare rate reduction

Clinical trials demonstrated meaningful reductions in lupus flare frequency and severity with Benlysta—allowing some patients to reduce corticosteroid use and better protect vital organs from the cumulative damage of repeated inflammatory episodes.

Frequently Asked Questions

What conditions does Benlysta treat?

Benlysta is FDA-approved for active systemic lupus erythematosus (SLE) in adults and children 5 years and older, and for active lupus nephritis — the kidney involvement that occurs in some lupus patients.

How is Benlysta given?

Benlysta is available as a monthly IV infusion given in a clinical setting, or as a weekly subcutaneous injection that can be self-administered at home after proper training. Your clinician will help determine the best option.

How quickly does Benlysta work for lupus?

Meaningful clinical response — reduced flare frequency and lower disease activity scores — typically develops over 6–12 months of consistent treatment. Earlier improvements in specific lab markers may be seen before this.

What side effects might I experience?

Nausea, diarrhea, infections, infusion reactions (for IV dosing), and injection site reactions (for subcutaneous dosing) are most common. Because Benlysta suppresses specific immune activity, susceptibility to certain infections increases.

Can Benlysta be used with other lupus medications?

Yes. Benlysta is typically used alongside standard lupus treatments including hydroxychloroquine, corticosteroids, and immunosuppressants — it's an add-on therapy for patients whose disease remains active despite standard treatment.
