What is minocycline?

Minocycline is a tetracycline antibiotic used to treat acne, certain respiratory infections, skin and soft tissue infections, sexually transmitted infections like chlamydia, and other susceptible bacterial infections. It's also used off-label for inflammatory conditions including rosacea and rheumatoid arthritis due to its anti-inflammatory properties. Minocycline works by binding to bacterial ribosomes and blocking protein synthesis. It has excellent tissue penetration and lipid solubility compared to other tetracyclines, making it effective for skin conditions and infections in hard-to-reach tissues. It's taken once or twice daily.

Bacterial ribosomal inhibition

Binds reversibly to the 30S ribosomal subunit, blocking aminoacyl-tRNA binding and preventing the elongation of bacterial protein chains — halting bacterial growth in a broad range of gram-positive and gram-negative organisms.

Tissue and skin penetration

Minocycline's high lipophilicity allows it to penetrate sebaceous glands, skin tissue, and other lipid-rich environments, which is why it is used for acne and skin infections.

Anti-inflammatory properties

Beyond antibacterial activity, minocycline inhibits neutrophil chemotaxis, matrix metalloproteinases, and pro-inflammatory cytokines — providing additional anti-inflammatory benefit relevant to acne, rosacea, and some autoimmune conditions.
